The "Big Bro" thing is based on the word "aniki", which is a very popular title to give to someone you see as superior to you, usually out of respect and sometimes with contempt. It's mostly used in yakuza settings with the men who work under a boss calling each other this term. When using "aniki" towards an ACTUAL older brother, it's a rude way of saying "oniisan."
