Yes, we need to unpack the data archive first (it is *.rgss3a for VX Ace. *.rgss2a for VX, etc) to apply our translation into the games. A tool like "RPG Maker Decrypter" is usualy used to extract archive data for RPG Maker games. But, RPG Maker MV has another story. Its single executable file you mentioned above is probably an Enigma Virtual Box. It is a software protection mechanism. The bad news is, it is not easy to extract them... and the good news is ... Translator++ has included an Enigma unpacker. So, we don't need to unpack these game to work with Translator++. But we still need the unpacked version to apply the tranlation though. You can use The Enigma unpacker to do this job. It is located at 'Translator++\3rdParty\EnigmaVBUnpacker.exe' or download it directly from its creator's website : https://lifeinhex.com/updated-enigma-virtualbox-unpacker-again/

Thank you, good programmer. I have one question. Some MV games have only executable(exe) file. (Probably it is a completely packed.) For these games, do I have to unpack to apply the translated data? This question also includes games made by VX ACE or VX.
